Sažetak

 

Uvod: Ki-67 je nuklearni protein koji je neophodan za proliferaciju ćelija. Vimentin se koristi kao marker za dokazivanje ćelija mezenhimalnog porekla. Ekspresija ovih markera može da doprinese boljoj interpretaciji proliferacije tumorskih ćelija i u ispitivanju njihove citoarhitektonike.
Cilj: Utvrditi Ki-67 proliferativni indeks i ekspresiju vimentina uz pomoć indirektne imunofluorescencije kod BHK-21/C13, MCF-7 i MRC-5 kontinuiranih ćelijskih linija.
Materijal i metode: Istraživanje je sprovedeno na kontinuiranim ćelijskim linijama BHK-21/C13, MCF-7 i MRC-5. Ćelijske linije su održavane u inkubatoru na 37 ºC, u atmosferi sa 100% vlažnosti i 5% CO2 tokom 48h. Sve ćelije su podvrgnute imunofluorescentnom bojenju na Ki-67 i vimentin antigen. Nakon toga, svaka ćelijska linija je fotografisana i fotografije su obrađene u Fiji softverskom programu. U okviru programa, određen je Ki-67 proliferativni indeks i jačina intenziteta imunofluorescencije za antitelo vimentin.
Rezultati: U sve tri ćelijske linije može se primetiti prisustvo Ki-67 antigena. Kod BHK-21/C13 proliferativni indeks je iznosio 97,01%; kod MCF-7 90,43%; kod MRC-5 90,58%. Intermedijarni filament vimentin je prisutan u citoplazmi svih ispitivanih ćelijskih linija. Jačina intenziteta imunofluorescencije za antitelo vimentin je pokazala najveće vrednosti kod BHK-21/C13, dok je najmanje vrednosti pokazala kod MCF-7.
Zaključak: Ki-67 predstavlja dobar pokazatelj ćelijske proliferacije, dok vimentin dobro opisuje citoarhitektoniku ispitivanih ćelijskih linija.

Abstract

 

Introduction: Ki-67 is a nuclear protein which is necessary for cell cycle proliferation, while vimentin is often used as a marker of mesenchymally-derived cells. Expression of these markers can contribute to better interpretation of tumour cell proliferation and cytoarchitecture during both normal development and metastatic progression.
Objectives: Determining Ki-67 proliferation index and vimentin expression by means of indirect immunofluorescence in BHK-21/C13, MCF-7 and MRC-5 continuous cell lines.
Methods: The research was conducted on continuous cell lines BHK-21/C13, MCF-7 and MRC-5. The cell lines were held in incubator on 37 ºC, in atmosphere with 100% humidity and 5% CO2 during the 48h. The cells were subjected to immunofluorescence for Ki-67 and vimentin antigen. Afterwards, every cell line was photographed and the photos were processed in Fiji software. Ki-67 proliferation index and the immunofluorescence intensity of vimentin antibody have been determined.
Results: Expression of Ki-67 antigen was noticed in all three cell lines. Proliferation index in BHK-21/C13 was 97.01%; in MCF-7 was 90.43%; in MRC-5 was 90.58%. Vimentin intermediate filament was present in the cytoplasm of all examined cell lines. Highest values of immunofluorescence intesity for vimentin antibody were shown in BHK-21/C13, while the lowest values were shown in MCF-7.
Conclusion: Ki-67 antigen represents a good cell proliferation indicator while vimentin refers well to cytoarchitecture of examined cell lines.
